- Avery Brooks (Benjamin Sisko), Nana Visitor (Kira Nerys), Rene Auberjonois (Odo), Alexander Siddig (Julian Bashir) and Armin Shimerman (Quark) were the only characters to appear in every episode since Colm Meaney (Miles O'Brien) often had to take time out, Cirroc Lofton (Jake Sisko) had a limit to how much time he could appear and the other regulars were limited term.
- Marc Alaimo (Gul Dukat), J.G. Hertzler (Vulcan captain/General Martok), Aron Eisenberg (Nog) and Mark Allen Shepherd () are the only actors, besides the regulars, to appear in both the first and the last episodes of the series.
- Rene Auberjonois, Avery Brooks, Michael Dorn, Alexander Siddig, Andrew Robinson, and TNG stars LeVar Burton and Jonathan Frakes also had a hand in directing episodes of the series. Dorn also directed in Enterprise. Robinson also directed in Voyager.
- Alexander Siddig (Julian Bashir) appeared in the opening credits by his birth name, Siddig el Fadil, for the first three seasons. He appeared as Alexander Siddig after he married co-star Nana Visitor (Kira Nerys), which placed their names together in the alphabetical cast credits. Siddig continued to be credited as Siddig el Fadil when he directed episodes.
- Trials and Tribble-ations was the first Star Trek episode of the TNG era to feature more than one of the TOS actors, and through superb special effects, features Colm Meaney being questioned severely by a young Captain Kirk regarding a fight aboard the K-5 space station. The resulting scene featured characters from three Star Trek series, Captain Kirk (Original Series), Miles O'Brien (Next Generation and DS9), and Doctor Bashir (DS9), appearing in the same scene.
- Liquidator Brunt, the unpleasant character from the Ferengi Commerce Authority, is played by Jeffrey Combs, who also features as the equally distasteful Weyoun, a Vorta administrator from the Dominion, who also has the privilege of being the character who has died the most often in the series. Combs also features as the Andorian Shran in the prequel series '.
- Morn is the only named character in the series who never utters a single line of dialogue. So great was the fan support for the silent character that a special episode was written for him in the sixth season: Who Mourns for Morn?, in which ironically, he still does not speak. Morn's name is an anagram of the character Norm from Cheers, who also spent seemingly all of his time sitting on his favorite bar stool and drinking.
- Regulars from other Star Trek series included Patrick Stewart, Jonathan Frakes, and Tim Russ. A second season episode, "Blood Oath," featured actors John Colicos, William Campbell and Michael Ansara as Kor, Koloth and Kang, reprising their roles as three of the first Klingons seen in the original Star Trek series.
- Academy Award Winner Louise Fletcher starred in a recurring role as the Bajoran spiritual leader Kai Winn.
- Other prominent celebrities seen in guest and recurring roles included Vanessa L. Williams, Lark Voorhies of Saved By The Bell, James Cromwell, Gabrielle Union, Iggy Pop, and James Darren.
