La Trobe has campuses in the Melbourne suburb of Bundoora (about 19,000 students), Bendigo (about 5000 students), Albury-Wodonga (about 2000 students) and minor campuses at Mildura, Shepparton, Beechworth and Mount Buller. Hundreds more overseas students undertake La Trobe courses in China, Hong Kong SAR, Vietnam, Singapore, Malaysia, Myanmar, Mongolia, Japan and Bhutan.

The University commenced operations in March 1967 as Melbourne's third university, after the University of Melbourne and Monash University, at what is now the Bundoora Campus. As a result of the Dawkins reforms to Australian higher education in the 1980s, La Trobe amalgamated with the Bendigo College of Advanced Education and Wodonga Institute of Advanced Education to acquire a range of regional campuses. In 1988 the Lincoln Institute for Health Sciences also merged with La Trobe. (The La Trobe Faculty of Health Sciences history document has more detail.)

The La Trobe University Medical Centre is based at the Bundoora campus of the university.

La Trobe University is named after Charles Joseph La Trobe. The University motto is "Qui cherche trouve" (French: "Who seeks, finds"). Professor Michael Osborne is the present Vice-Chancellor and President of La Trobe University.
