The Derwent Valley Line is a railway line from Derby to Matlock in Derbyshire.

The line follows the Midland Main Line as far as Ambergate Junction, which is south of Ambergate railway station, originally built by the North Midland Railway. The line then continues to Matlock, following the course of the River Derwent along what was originally the Manchester, Buxton, Matlock and Midlands Junction Railway.

- Former mineral branch line diverges to Wirksworth. This route has now closed, but is being reopened as the Ecclesbourne Valley Railway heritage railway.

- Route diverges away from the Midland Main Line just to the south of Ambergate station.

- Matlock Bath
- Matlock
- The route through to Buxton closed in 1968. Part of this line has now been preserved as the Peak Rail.
Local passenger services are operated by Central Trains between Derby and Matlock. Services are approximately hourly, but with some gaps. The line is also used by Midland Mainline and Virgin Trains between Derby and Ambergate Junction.
